Emerging methods for exosome purification and process monitoring

Pete Gagnon, CSO of BIA had a presentation at ISBioTech 9th Spring Meeting entitled "Emerging Methods for Exosome Purification and Process Monitoring«. Also, on 22nd of March BioProcess International eBook covering the same theme was released. It is well known that  exosomes bring a new set of challenges to purification; some in common with proteins, some in common with virus particles, but many unique to exosomes. Case study data, shown in both presentation and the eBook, illustrated the application of hydrogen bond chromatography and MALS to purification of vesicles from plasma and cell culture harvest.

Comparison of fluorescence and multi-angle light scattering for rapid  quantitation of bacteriophage

 
One of the handicaps of working with bacteriophages is the long duration required to perform plaque assays. Plaque assays also impose questions about accuracy and
precision relative to the scale and experience of the persons performing and interpreting them. This poster presents a pair of high precision, high accuracy chromatography-based assays that permit determination of phage concentration in less than 1 hour.

Estimation of empty and full AAV10 particle ratio using the multi-angle light scattering detector

 
Serotype 10 adeno-associated virus (AAVrh_10mCherry) was analysed on the PATfix™ HPLC system with the  CIMac™ AAV full/empty analytical column to estimate the ratio of empty and full AAV particles based on the peak area of the chromatogram given with three different detectors. AAV included a protein capsid containing single stranded DNA. CIMac™ AAV column consisted of a strong anion exchanger with QA chemistry (quaternary amine).

Two step purification process for bacteriophages with CIM® monoliths

 
In bacteriophage research and therapy, most applications ask for highly purified phage suspensions as such it is crucial to reduce proteins, endotoxins, DNA and other contaminants. In this study we present efficient two-step chromatographic purification method with binding phages to a stationary phase - monoliths. The aim of the study was to develop robust, fast and effective virus purification platform that can be used for several types of bacteriophages for any application.
