Welcome to the Summer 2020 FiberCell Expressions Newsletter...

It seems like just last week I was returning from a trip to Japan, on March 1. Since then so much has happened, but the time has just flown by. Many universities remain closed, many companies are still having employees work from home. Researchers in Asia and Europe have started to get back to work, and I can see in the US that researchers are coming up for air and getting back to work, and there is much important work to be done. FiberCell Systems is involved in many aspects of COVID-19 research from production of diagnostic antibodies, experimental exosome therapies, and pk/pd studies of anti-viral drugs. I remain cautiously optimistic about what is to come.
 

First time user of FiberCell Bioreactor produces nearly 2 grams of antibody
 
Here is some nice data that we got recently from Dr. Natalia Bogdanova at the Texas Medical Center.  This is interesting for two reasons 1) this was her first time working with a hollow fiber bioreactor and 2) some interesting observations regarding insulin supplementation of the media. Insulin is generally not required in high density cell culture. The glucose rates were greater than 3 grams per day, much too high, and antibody production was reduced. Once the insulin was removed production and growth rates were standard for FiberCell. Dr. Bogdanova interacted with us on a daily basis to get her bioreactor under control and produced 1.9 grams of antibody in a volume of 1200 mL, (1.59 mg/mL), and  consumed 37 liters total of DMEM/CDM-HD. Note how clean the gel is, with the antibody of interest the major protein component. It was a little tricky at first, but I love a happy ending.

www.hollowfiberinfectionmodel.com is a new website that is dedicated specifically to sharing results, data, papers and protocols for Pk/Pd studies on antibiotics and also antiviral drugs. It is intended to serve as a resource for both new users and for experienced users of the hollow fiber infection model. There is also a forum where you can post questions and share experiences. You will also find equipment lists and training videos.

FiberCell Systems introduces new PVDF cartridges for Pk/Pd studies
 

 
PVDF (polyvinylidene difluoride) is a well known low protein binding, low non-specific binding polymer. There are some antibiotic drugs, particularly those used as anti-tuberculosis drugs, or some of the newer antibiotics that are very polar in nature and can require significant amounts of solvent to get them into solution. These new cartridges are the same size and volume as the C2011, have higher solvent resistance than the polysulfone fibers, and much higher flux rates than the cellulosic fibers. The pore size is .03 um and have been validated for complete retention of mycobacterium such as TB.

FiberCell Systems Poster at ISEV describes higher bioactivity exosomes from Human Amniotic Stem Cells


 
ISEV 2020, the virtual meeting, has come and gone. There were over 1200 “attendees” and though it missed the socialization and face to face interactions of meetings in the past, it was nice to be able to save talks and view them on your own schedule so nothing was missed. FiberCell Systems presented a poster in conjunction with Dr. Laura Perin from UCLA Children’s Hospital. She used our smallest cartridge to produce extracellular vesicles from her human amniotic stem cells that are used as a therapy for Alport’s disease, a genetic abnormality of the kidney. The 3-D capillary bioreactor produced as many EVs as 25 petri dishes, every day, and demonstrated enhanced bioactivity when compared to 2-D EVs.

FiberCell Laboratories is Open



Like many researchers we shut down our laboratory the second week of March. We used that time to move to a new space and upgrade our capabilities. We are back up and running with a few projects ongoing but still have the time and space for some more projects. Maybe you would like to produce an antibody or a recombinant protein? Perform some pilot experiments for a grant or other project?  Feel free to drop us a line. We are always happy to discuss how 3-D capillary bioreactors can be a more in vivo like way, and a better way to grow cells.
